Roger Federer defeated Andre Agassi 6–3, 2–6, 7–6^(7–1), 6–1 in the final to win the Men's Singles tennis title at the 2005 US Open.

This was the first US Open that future champions Novak Djokovic and Andy Murray competed in the main draw.

Noteworthy in this tournament was a strong performance by the American men, despite former champion and world #1 Andy Roddick losing in the first round.

Wildcard James Blake made the quarterfinal, upsetting #2 Rafael Nadal in the third round before eventually losing to Agassi in a five-set match.

Unheralded American Robby Ginepri, whose run was enhanced by Roddick's early defeat, made it all the way to the semi-final and lost to Agassi in another five- setter.
